How transfer music?

Just installed Poweramp on Galaxy10.

Plugged the phone in computer and turned on Android file transfer

How can I transfer music then?

Transferring music to your phone is nothing to do with Poweramp, as an audio player PA just plays what you have already placed on the device.

When you have connected your phone to your computer via a USB cable, open a PC file explorer window to your phone's storage (internal or SD Card) and simply drag whatever music folders you want from your PC's hard drive window over to the phone's window.

You might find it quicker if copying a lot of files to simply remove the SD Card from the phone and plug it into your computer (using a card slot or USB adapter). Direct copying that way is always going to be quicker than using the phone's firmware as a link. 

By "Galaxy10", do you mean an old Samsung S10 or a newer A10?
